Abstract
Capillary isoelectric focusing (CIEF) was carried out using a commercial capillary isotachophoretic analyzer, the LKB 2127 Tachopor. The unmodified instrument was found to be unsuitable for fast focusing, due to non-optimized boundary conditions at the capillary ends, the length of the separation tube, and insufficient cooling of the focusing trough. Simple column changes were implemented, resulting in significantly improved applicability of the instrument for CIEF. The impact of these modifications was examined via the analysis of protein mixtures in both simple buffers and in synthetic carrier ampholytes.
