Abstract
The selective loading of Kinsein-powered molecular shuttles with protein cargo and its application in biosensing is discussed. The initial coating of the Kinsein-bound, biotinylated microtubules with streptavidin has to fill the available biotin binding sites to avoid unwanted cross-linking of microtubules with each other. It permits the selective capture of proteins, and the subsequent detection of the capture event in double-antibody sandwich geometry. It enables the loading of all cargo types against which antibodies can be raised.
